1. Place one (1) Iron Plate on the top-left Transport Belt of the Transport Belts that are connected to the Circuit Network (see image), preferably in the inner portion. (Press Z to drop item in cursor) This Iron Plate will support the latch system. 2. Supply the left Transport Belt bus with U-238 on the LEFT SIDE ONLY. If both sides are used the system could jam.
3. Place 40 OR preferably 80 U-235 within the Centrifuge.
(Optional) 4. Supply the Centrifuge with Modules if desired.
The mechanism should now continue to run on its own.

The base mechanism only costs 103 Iron Plates, 42.5 Copper Plates, and 8 Steel Plates. The Centrifuge, however, costs 400 Iron Plates (and 10 Iron Ore), 500 Copper Plates, 50 Steel Plates, 100 Stone, 1k Water, and 200 Plastic Bars. So, the mechanism can be considered relatively dirt-cheap.
