How can I pick something into my hand? For example there are some walls in my inventory and I want to build them. What button do I need to press to do so? I can't find that point in the key assignments.

So basically placing is by taking it into your "hand" and press the left mouse button.

I know all that keys, but what I'm looking for is a way to directly pick up an item out of my inventory to use it

The point is I don't know thebutoon how I get the item into my hand. (Except of the express bar) I tried already like everything and still it doesn't work

So pick up an item from your inventory (press E, that's what stands in the key-controls in game) then you click on one of the icons and you have it in the hand and place that item on ground with the left mouse (also defined in the key control). You can see that working also on most let's play videos on YouTube, which I recommend very strong, cause that is a very basic level of support, which this board is in my eyes not thought for.

The reason to point to this key control is, that you may accidentally miss-configured it and it explains all the other controls (there are many and you are now at the first very basic) more or less well.

Bu thats the problem, if I press left click on the item nothing happens and if I press right click i get 50% of it in my hand

If you can't pick up a full stack with left mouse button, your key may be unbound.

And what is the name of this function in the key assignment option

In the controls options, go to the inventory tab. There's everything. "Pick up item" should be bound to the left mouse button. It says so in singular form, but it's supposed to pick up the whole stack.
